The P100 and P300.
The P100 is a positive peak in the grand average response-time-wave form observed at around 100 msec following visual stimulus onset in individuals who can monitor visual displays for feedback information. This response reflects the visual system's ability to extract information from the stimulus and communicate it to higher level cognitive structures. The P100 is sensitive to stimulus features and response requirements. Changes in the P100 reflect changes in the stimulus and information required to respond. The P300 is a negative peak in the grand average response-time-wave form, occurring at around 300 msec following visual stimulus onset. The P300 reflects the processes of stimulus classification, response selection, and decision making. The P300 is sensitive to stimulus features and response requirements. The P100 and P300 are involved in both central and peripheral processing of the visual stimulus. The P100 is an early visual event which occurs before the P300. The P300 occurs later in the stimulus processing, reflecting the processes of decision making, response selection, and execution. We have evaluated the use of minisequencing for the detection and mapping of VH gene rearrangements in the human IgG heavy chain. To facilitate minisequencing, we have constructed a semiquantitative PCR strategy for the amplification of target sequences by using a vector that contains a unique restriction site immediately downstream of the target sequence. The relative size of the products from this PCR enables quantitative comparison with respect to a size-standard series of target sequences. We show that minisequencing can detect the actual rearrangements of VH gene segments from a recombinant myeloma cell line producing an anti-CD19 antibody.
